Bovine response to lipoarabinomannan vaccination and challenge with Mycobacterium paratuberculosis BJM

This study aimed to evaluate the immune response in bovines following immunization with a mycobaterial Lipoarabinomannan extract (LAMe) and the effect of Map challenge. LAMe vaccine induced specific antibody levels that diminished after the challenge and affected Map excretion at least for 100 days thereafter.

In Chile, information about Map isolation from both domestic ruminant and wildlife species has been accumulating, but it has to be extended to other species. Detection of Mycobacterium avium subsp. paratuberculosis in bovine milk from the state of Pernambuco, Brazil BJM

Abstract The aim of this study was to detect the IS900 region of Mycobacterium avium subsp. paratuberculosis (MAP) in bovine milk samples using real-time polymerase chain reaction (qPCR) and conventional PCR, and to study the agreement between these tests. A total of 121 bovine milk samples were collected from herds considered positive for MAP, from the State of Pernambuco, Brazil. MAP DNA was detected in 20 samples (16.5%) using conventional PCR and in 34 samples (28.1%) using qPCR. MAP DNA was detected in all of the 6 animal farms studied. La paratuberculosis es una enfermedad granulomatosa crónica intestinal, causada por Mycobacterium avium subespecie paratuberculosis, que afecta a los rumiantes. El objetivo del trabajo fue caracterizar la respuesta inmune humoral sistémica y local de bovinos infectados. Se estudiaron once bovinos, seis infectados (asintomáticos, sintomáticos y terminales) y cinco controles. First identification of Mycobacterium avium paratuberculosis sheep strain in Argentina BJM

We here identified for the first time the presence of Mycobacterium avium paratuberculosis (MAP) sheep (S) strain in Argentina. IS900 polymerase chain reaction (PCR) was positive. The S strain was compared with MAP cattle (C) strains by using IS1311 PCR-restriction endonuclease analysis (PCR-REA), multiplex PCR and restriction fragment length polymorphism (RFLP) analysis.

First isolation of Mycobacterium avium subsp paratuberculosis from commercial pasteurized milk in Argentina BJM

Mycobacterium avium subsp paratuberculosis was isolated from two out of seventy samples (2.86 %) of pasteurized and ultra-pasteurized milk. The isolates were positives to IS900 PCR and showed a C17 RFLP pattern, the most prevalent in Argentina. The present study is the first report of Mycobacterium avium subsp paratuberculosis culture from pasteurized milk in Argentina.

Descreve-se o primeiro caso de paratuberculose em veado pudu-do-sul (Pudu pudu). O animal foi encontrado na cidade de Concepción, centro sul do Chile, em péssimas condições e com sinais claros de diarréia. Internamente apresentava séria atrofia de gordura no coração, lesões histológicas nos pulmões, fígado, baço e rins, e injúrias intestinais. Diferentes amostras foram preparadas com Ziehl-Neelsen, e os linfonodos mesentéricos presentes no íleo foram fortemente positivos para a bactéria resistente ao ácido.

Improvement of an in-house ELISA for bovine paratuberculosis serology in Brazil BJM

Paratuberculosis (Johne's disease) is a chronic enteritis in cattle caused by infection with Mycobacterium avium subsp. paratuberculosis (Map). In Brazil, few reports describe the isolation of the organism. The gold standard test for Map is its isolation from tissues or feces. Moreover, bacterial growth is slow and test results are available only after three to five months of incubation Furthermore, shedding of bacilli at levels detectable by fecal culture is irregular and does not occur during the early stages of infection, which compromises the sensitivity of this methodology. Interference of tuberculosis on the performance of ELISAs used in the diagnosis of paratuberculosis in cattle BJM

Forty-four cows from five herds infected with tuberculosis (TB) and without paratuberculosis (PTB), and 21 cows from a herd without either infection were studied. The cattle presented concordant results in both the skin test and gamma-interferon assay for TB and two commercial ELISAs for PTB. Animals were divided according to TB test results into Group A with 28 TB-infected animals, Group B with 16 TB-negative animals from infected herds, and Group C with 21 TB-negative cows from a tuberculosis-free herd (which were used as controls). Actualmente se vincula al Mycobacterium avium subsp. paratuberculosis como potencial agente etiológico implicado en la enfermedad de Crohn en humanos. Una de las vías de ingreso sería a través de la ingestión de leche contaminada. El objetivo fue evaluar la presencia de Map en leche comercial homogeneizada y ultrapasteurizada para consumo humano en supermercados de la ciudad de Río Cuarto, Córdoba, Argentina. Los camélidos sudamericanos (CS) incluyen cuatro especies, guanaco, vicuña, alpaca y llama (Lama glama). LAMP technology: Rapid identification of Brucella and Mycobacterium avium subsp. paratuberculosis BJM

In this study, we developed new sets of primers to detect Brucella spp. and M. avium subsp. paratuberculosis (MAP) through isothermal amplification. We selected a previously well-characterized target gene, bscp31, specific for Brucella spp. and IS900 for MAP. The limits of detection using the loop-mediated isothermal amplification (LAMP) protocols described herein were similar to those of conventional PCR targeting the same sequences. Hydroxynaphtol blue and SYBR GreenTM allowed direct naked-eye detection with identical sensitivity as agarose gel electrophoresis. La paratuberculosis es una enfermedad que afecta al ganado vacuno cuyo agente etiológico es el Mycobacterium avium subespecie paratuberculosis. El LAM es el principal componente antigénico de superficie de las micobacterias, y se lo considera de relevancia en la patogenia de las enfermedades que éstas causan. Un extracto enriquecido en LAM fue obtenido a partir de un cultivo de Mycobacterium spp. y empleado para inocular terneros. Multiple-locus variable number-tandem repeat analysis (MLVA) of Mycobacterium avium subspecies paratuberculosis (MAP) isolates may contribute to the knowledge of strain diversity in Argentina. Although the diversity of MAP has been previously investigated in Argentina using IS900-RFLP, a small number of isolates were employed, and a low discriminative power was reached. It is caused by Mycobacterium avium subsp. paratuberculosis (MAP) that may also be involved in some human diseases such as Crohn's disease, type 1 diabetes, sarcoidosis, multiple sclerosis, and Hashimoto's thyroiditis. Paratuberculosis is a chronic enteritis that affects ruminants and is caused by Mycobacterium avium paratuberculosis (Map). The disease is worldwide spread and causes important economic losses. In Brazil, the bacillus was isolated in the south and northeast regions of the country and in Rio de Janeiro, but there are no enough epidemiological studies about its occurrence. To study the deficiency of minerals and its relationship with Paratuberculosis, blood, serum, and fecal samples were obtained from 75 adult bovines without clinical symptoms of the disease and from two bovines with clinical symptoms of the disease, from two beef herds with a previous history of Paratuberculosis in the Province of Buenos Aires, Argentina. Serum samples were processed by ELISA and feces were cultured in Herrolds medium. Copper, zinc and iron in serum were quantified by spectrophotometry and selenium was measured by the activity of glutathione peroxidase.
